Transgenic potatoes StSDDhpi exhibit an increase in stomata density. Most probably this has an effect on gas and water exchange of the plants with the atmosphere. The genetically modified plants displayed increased heat tolerance and since this influences photosynthetic activity the increase in stomata density is thought to also increases the yield of potato tubers. To achieve this increase in stomata density the gene stomatal density and distribution 1 (sdd1) was silenced via RNAi since a sdd1 knock-out mutant of Arabidopsis thaliana demonstrated that sdd1 is a negative regulator of stomata development.

The term “Recipient organism” refers to an organism (either already modified or non-modified) that was subjected to genetic modification, whereas “Parental organisms” refers to those that were involved in cross breeding or cell fusion.

The inserted T-DNA consists of several cassettes: First, a RNAi construct that is build of a StSDD1 gene fragment in sense- and antisense orientation separated by the pdk intron 3 promotes the generation of double stranded RNA (dsRNA) in a hairpin conformation. This dsRNA targets the endogenous SDD1 transcript and therefore mediates post transcriptional gene silencing. The RNAi construct is controlled by the CaMV 35S promoter and the ocs terminator.
Second, as a selection marker system nptII gene is used whose gene product aminoglycoside 3-phosphotransferase II confers resistance to aminoglycoside antibiotics (kanamycin, neomycin, gentenicin). NptII is controlled by the nos promoter and -terminator.
Furthermore, sequence fragments of the lacI gene, the lacZ promoter and the lacZ gene of Escherichia coli are encoded by the inserted DNA but are not functional in plants.
